Novel Antimicrobials From African Biodiversity: A Drug Discovery Renaissance
African researchers are harnessing the continent's unique biodiversity to discover novel antimicrobial compounds. Three promising molecules derived from African soil microorganisms are entering preclinical trials.
The global antimicrobial resistance crisis is compounded by a drying pipeline for new antibiotic discovery. However, an emerging movement of African researchers is reversing this trend by harnessing the continent's exceptional and largely unexplored biodiversity to identify novel antimicrobial compounds. The AfricaBioAntibiotic project, coordinated by the University of Pretoria and the H3D Institute at the University of Cape Town, is at the forefront of this renaissance.
The project has assembled a biobank of over 50,000 microorganisms isolated from soil samples, medicinal plants, and extreme environments collected across 20 African countries. High-throughput screening has identified 127 extracts with significant antimicrobial activity, including three particularly promising molecules that target novel mechanisms of action against multidrug-resistant Gram-negative bacteria.
Professor Kelly Chibale, director of the H3D Institute and a pioneer in African drug discovery, stated that "African biodiversity holds a treasure trove of antimicrobial molecules that the world has yet to explore. Our goal is to transform this natural wealth into therapeutic solutions while ensuring that benefits flow back to the communities and countries of origin." He emphasized the importance of the Nagoya Protocol for ensuring equitable benefit sharing.
The three candidate molecules — named AfriMycin-1, AfriMycin-2, and AfriMycin-3 — have shown potent activity against carbapenem-resistant Klebsiella pneumoniae and pandrug-resistant Acinetobacter baumannii in vitro. Preclinical trials, funded by GARDP and the Wellcome Trust at $15 million, will begin in the third quarter of 2026 at GLP-certified facilities in Cape Town and Nairobi.
